Directions:
- In a heatproof dish or bowl stir 1,2 gr (1/2 tbsp.) of agar powder into 60 ml (1/4 cup) of hot water. The size of each Petri dish is 10 cm (3.9") in diameter. Multiply these quantities by the amount of Petri dishes you plan on using. It means that for 10 dishes you'll need 5 tbsp. of agar and 600 ml (2,5 cup) of water.
- Boil this mixture for 1 minute until the agar dissolves completely.
- Allow the agar solution to cool down for several minutes before proceeding.
- Pour the layer of agar solution in each Petri dish. Make sure to open and close the lid quickly to prevent any airborne bacteria from contaminating the experiment.
- Set the Petri dishes aside for 30 minutes to 2 hours, until the agar solution cools down and hardens.
- Once the agar solution has hardened and the Petri dishes are at room temperature, you are ready to introduce the bacteria through direct contact. Refrigerate the Petri dishes if not used immediately.
- Secure each Petri dish with masking tape. Don't forget to mark in a discrete way to whom the sample belongs.
- Place and incubate the Petri dishes in a warm, dark place. Remember to store the dishes upside down, so the bacterial growth remains undisturbed by any water droplets. The ideal temperature for growing bacteria is between 20-37 degrees C (70-98 degrees F). Leave the bacteria to develop for 4-7 days.
- When ready, hold each Petri dish with your thumb and index finger. Make a mark where you place your thumb with a tape or pen marker. This will be the equivalent of a “mug holder” for following readings. It's best to handle Petri dishes with rubber gloves in case some of the bacteria might be hazardous.
- Interpret the bacterial shapes according to a tea-cup fortune-telling method. Visualize the bacterial constellations as tea leaves, and the shape of the Petri dish as the oval shape of a tea cup. There are two major factors you should pay attention to:
